不结球白菜下胚轴和子叶离体培养再生植株 被引量:14

摘要 以不结球白菜恢复系91H秋-21种子的无菌苗子叶、下胚轴和地方品种苏州青的子叶为外植体,在附加适当外源激素的改良MS培养基上诱导产生愈伤组织,继代培养后再分化,获得再生植株。在愈伤组织继代培养基中添加适量的KT,有利于绿苗分化。在分化培养基中,不同种类的细胞分裂素对再生绿苗的效果有显著差异,CPPU最好,植株再生频率可达10.0%;ZT居中,为3.8%;BA较差,为1.1%;使用KT未得到再生植株。 Calli were induced from cotyledons and hypocotyls of nonheading Chinese cabbage on a modified MS medium (B5 organic ingredients) with 1.0 mg/L 2, 4-D and 0.5 mg/L KT.The calli obtained were subcultured on a medium with 0. 2mg/L 2, 4-D and 0.5 mg/L KT for 3- 4 weeks,then they were transferred to a fresh medium with cytokinins for redifferentiation.Plantlets were obtained from calli of two cultivars. Shoots could not be produced directly from these calli without subculture. KT suplement in the subculture medium was useful to shoot regeneration. Several kinds of cytokinins showed different effects on shoot regeneration in the differentiation medium:CPPU had the best effect, BA had a modest effect,and the effect of ZT was between CPPU and BA.
