摘要
目的:研究三羟基异黄酮在体外对结肠癌细胞株HT-29的作用。方法:三羟基异黄酮作用于10%FCS的PRMI-1640液培养HT-29后,MTT法测定吸光度OD;流式细胞仪测定细胞DNA含量,做细胞周期分析,计算凋亡率,观察其对细胞增殖和凋亡的影响。结果:不同浓度的三羟基异黄酮在不同生长时间均对HT-29细胞增殖有抑制作用(P<0.05);三羟基异黄酮作用于HT-29细胞后,可以观察到G1峰前出现亚G1峰高于对照组(P<0.01),对照组凋亡率为2.54±0.46%,实验组为7.72±0.55%。结论:三羟基异黄酮可以抑制HT-29的增殖,促进其凋亡。
Objective : To study the effection of 3 - droxide isoflavone on colon cancer cell HT - 29 in vitro. Methods : Cultures HT- 29 by 10% FCS PRMI - 1640 and 3 - dioxide isoflavone, measure OD by MIT and DNA content by FCM, analyses cell cycle and calculates apoptosis rate. Results: HT - 29 cell proliferation could be inhabited by different concentration of 3 -droxide isoflavone at different accretion stages ( P 〈 0.05) ; The effected cell appeared sub - G1 peak before G1 peak (apoptosis rate: 2.54±0.46% vs7.72±0.55%). Conclusion : 3 - droxide isoflavone can inhabit of proliferation and induce apoptosis of HT- 29.
