摘要 通过生理、生化特性,抗生素敏感性,转化实验及质粒的提取和鉴定等实验,对嗜酸乳杆菌ATCC4356和乳酸乳球菌NZ9000作为宿主系统的可行性进行研究。结果表明,乳酸乳球菌NZ9000和嗜酸乳杆菌ATCC4356均可作为宿主系统,表达pLP352质粒(氯霉素抗性质粒,可在大肠杆菌和乳酸菌中穿梭)。研究结果,在基因工程上应用抗病基因在乳酸菌中的表达奠定了基础,证实了乳酸菌作为生物安全级别的宿主系统的可行性。 In the study, we verified physiological and biological characteristics, antibiotic resistance, transformation and extract plasmid and identification etc. Test to Lactobacillus, and we identified the feasibility of Lactococcus lastic NZ9000 and Lactobacillus acidophilus ATCC4356 as host system. We can get a conclusion that Lactococcus lastic NZ9000 and Lactobacillus acidophilus ATCC4356 could be as the host system and express the shuttle expression plasmid pLP352 (chloromycetin resistance) between E. coli. and lactic acid bacteria. Our study have laid the ground work for some useful combat disease gene expressed in the lactic acid bacteria, and testified the feasibility of Lactobacillus as biological safe grade host system.
