摘要 目的建立测定犬血清中盐酸伪麻黄碱的方法。方法采用高效液相色谱法,血清样品用乙醚提取,以ODSC18为固定相,甲醇∶乙腈∶磷酸盐缓冲液(pH3.5)=30∶10∶60为流动相,UV210nm紫外检测。结果在0.031252.0mg.L-1浓度范围线性良好,最小检测浓度为0.03125mg.L-1,最低检测限为0.625ng。其日内差(n=5)及日间差(n=5)RSD分别小于7.5%和7.6%,相对回收率(n=5)大于91.0%。结论方法简便灵敏,为盐酸伪麻黄碱的药代动力学研究提供了检测方法。 Aim To establish a method for determination of pseudoephedrine hydrochloride in dog serum. Methods Pseudoephedrine was extracted with ethyl ether,and then measured by RP-HPLC using a ODS C18 coWlumn asstationarv phase and methanol-acetonitrilephosphate buffer solution(pH 3.5) (30: 10: 60)as mobile phase with ultraviolet absorption at 210nm. Results The method proved to be linear in the range of 0.03125-2.0 mg· L^-1 and the lower limit of detection of pseudoephedrine hydrochloride in serum was 0.03125 mg·L^-1. The RSD was under 7.5% (n=5 ) and 7.6% (n=5 ) for inter-assay and intra-assay respectively, and the retative recovery rate was superior to 91.0%. Conclusion This method is sensitive,specific,simple and validated for use in pharmacokinetic study.
