摘要
目的:探讨血管紧张素Ⅱ在不同浓度和不同作用时间下,对血管平滑肌细胞凋亡的作用及卡托普利和氯沙坦对其影响。方法:采用流式细胞技术测定了在血管紧张素Ⅱ不同浓度和不同作用时间下,血管平滑肌细胞凋亡率的变化和卡托普利、氯沙坦及二者舍用对其影响。结果:血管紧张素Ⅱ中低剂量对血管平滑肌细胞凋亡无明显作用,大剂量有诱导凋亡的作用;单用氯沙坦对细胞凋亡无明显作用,卡托普利有一定的作用,二者合用时明显增加凋亡率。结论:血管紧张素Ⅱ具有促进血管平滑肌细胞凋亡作用,大剂量作用尤为明显;合用卡托普利和氯沙坦可明显增加平滑肌细胞的凋亡作用。
Objective: To study the effect of captopril and Iosartan on apoptosis ratio induced by angiotensin Ⅱ in different concentrations and different action time in vascular smooth muscle cells. Method : The flow cytometer was used to measure the changes of apoptosis ratio induced by angiotensin Ⅱ in different concentrations and different action time and to observe the influence by captopril and losartan on it. Result: The apoptosis ratio was induced and increased by angiotensin Ⅱ, especially in high concentrations. The pro - apoptotic effect was not established only by losartan. Captopril had some effect on the apoptosis ratio; however, the apoptosis was significantly increased when cells were pretreated with the combination of losartan and captopril. Conclusion: The apoptosis ratio can be induced and increased by angiotensin Ⅱ, especially in high concentrations. The combination of losartan and captopril may have the inhibition effect on AngⅡ and apoptosis in vascular endothelial cells.
