摘要
目的优选出虎杖中白藜芦醇的最佳提取奈件。方法以提取溶剂乙醇浓度、乙醇用量、提取时间、提取次数为4个考察因素,以干膏得率、白藜芦醇含量为考察指标,运用Doehlert设计、多重响应的渴求函数优化法对虎杖中白藜芦醇的提取条件进行优化。结果优选的提取条件为以7倍量95%乙醇回流提取2次,每次1h。结论应用Doehlert设计、渴求函数的响应曲面优化法具有使用方便、预测性好的特点,值得推广应用。
OBJECTIVE To optimize the extraction process of resveratrol from Polygonum cuspidatum Sieb.et Zucc.METHODS A doehlert design matrix, which allowed a sequential response surface methodology to be performed, was used for the optimization of the extracting condition, Four independent variables, i.e., ethanol concentration, ethanol volume and extracting times were investigated. Yields, as well as resveratrol content, were estabhshed as responses. And the desirabihty function was finally used to simultaneously optimize the two considered responses. RESULTS The optimum extraction conditions were as follows: refluxing and extacting with 7-fold 95 % alcohol for 2 times and 1 h for each time. CONCLUSION The doehlert design and desirability function, which allow a sequential response surface methodology,are both good methods in process optimization.
