摘要
目的:研究不同培养条件对天山雪莲愈伤组织生长及总黄酮产生的影响,探讨组织培养生产总黄酮的可能性。方法:通过脱分化培养获得天山雪莲愈伤组织,并对该组织含有的总黄酮进行测定。结果:在MS+NAA+6-BA培养基上培养10~15d,能获得紫红色愈伤组织,且总黄酮含量较高;温度梯度、NH4NO3浓度、培养基和培养时间对愈伤组织的生长和总黄酮产生均有显著影响。结果:天山雪莲愈伤组织培养条件对总黄酮产生有影响。
Objective: The effects of different conditions on the callus growth of Saussurea involucrata Kar.et Kir. and the production of total flavonoids were reported in this paper. Besides the possibility of producing total flavones by means of tissue culture was investigated. Method: We obtained the callus of S. involucrata by way of dedifferentiation culture and determined the contents of total flavones. Result: After being cultured for 10- 15 days on MS+NAA+6-BA media, the callus turned vivid amaranth and had a high content of total flavonoids. The contents increased to 1.47 times after being cultured under gradient temperatures. Characters of the callus, media,culture time, culture temperature and the concentration of NH4NO3 all had remarkable effects respectively on the growth of callus and the production of total flavonoids. Conclusion: The cultural conditions were the maini mpact factors affecting the total flavonoids accumulation.
