摘要
目的调查四川地区人群HLADRB1位点等位基因多态性,获得完整准确的遗传学数据。方法采用单管PCR扩增方法建立高效的HLADRB1基因测序分型(SBT)技术对180名健康个体进行HLADRB1基因型检测。结果共检出39种HLADRB1等位基因,其中等位基因HLADRB10901的频率最高,HLADRB11101的频率次之。结论提供了四川地区汉族人群HLADRB1位点等位基因频率,证实了单管扩增测序分型方法快速准确的优点。
Objective To determine HLA-DRB1 genetic polymorphism in Sichuan Han population and establish ethnic gene database of HLA-DRB1. Methods A set of 7 group-specific exonic 5' amplification primers and a single generic 3' primer were included together in a single PCR mix to facilitate a single PCR amplification per sample for HLA-DRB1 typing. 180 Sichuan Han healthy subjects were investigated. Results 39 alleles were identified in Sichuan Han population with the most frequent allele being HLA-DRB 1 * 0901, followed by HLA-DRB1 * 1101. Conclusion Data of HLA-DRB1 polymorphism in Sichuan Han Ethnic Group will contribute much to the improvement of research on disease association and basic immunology.
