摘要
目的探讨单纯疱疹病毒2型(herpes simp lex virus type 2,HSV-2)直接感染诱导小鼠阴道黏膜的细胞凋亡情况与Fas,Bc l-2蛋白水平的关系。方法建立HSV-2阴道感染小鼠模型。用TUNEL染色研究HSV-2诱导阴道上皮细胞凋亡,检测Fas,Bc l-2蛋白的水平,分析其与细胞凋亡的相关关系。结果22份标本有凋亡细胞阳性表达,并同时伴有炎症及坏死。Fas蛋白阳性率高(24/24),Bc l-2蛋白阳性率低(2/24)。结论HSV-2直接感染小鼠阴道后可同时诱导阴道上皮细胞凋亡及炎症坏死,Fas蛋白呈高水平,与凋亡细胞指数呈正相关(r=0.9660,P<0.05),提示Fas对HSV-2诱导的黏膜细胞凋亡起上调作用。
Objective To study the apoptosis induced by HSV-2 in the mice infected directly intravagina mucous membrane and analysis the relationship between apoptosis and level of Fas and Bcl-2 proteins, Methods The model mice of positive HSV-2 infection was established for demonstrating apoptosis with TUNEL staining and detecting Fas protein and Bcl-2 protein,and analyzed the relationship between apoptosis and protein expressions. Results In 22 infected specimens, positive reaction of the apoptosis and the necrotic areas and inflammatory reaction in epithelia could be found at the same time, the ratio of Fas protein was high (24/24), and Bcl-2 was low (2/24), Conclusion The apoptosis and the necrotic and inflammatory in the vaginal epithelial of mice model could be induced by infection of HSV-2 at the same time, The correlation between the AI ( apoptosis index ) and Fas protein was positive( r = 0. 9660 ,P 〈 0. 05), This suggested that Fas protein can up-regulate the apoptosis,
