人表皮干细胞体外分选和培养对构建组织工程皮肤种子细胞的可能性 被引量:5

摘要 目的:探讨人表皮干细胞在体外分选和培养的可能性,为构建组织工程皮肤寻找理想的种子细胞。方法:实验于2002-01/12在中山大学基础医学院动物中心完成。根据表皮干细胞与Ⅳ型胶原蛋白的高黏附的特性,运用黏附实验将干细胞从培养在鼠Ⅳ型胶原包被的培养瓶的表皮细胞中分选出来,并从形态学、超微结构及免疫组化等方面进行了鉴定。结果:分选出来培养的细胞符合表皮干细胞的特征,说明分选培养的细胞是表皮干细胞。结论:预先在体外对表皮干细胞进行分选和培养是可行的,该细胞群有望成为构建组织工程皮肤的种子细胞,为下一步应用表皮干细胞构建组织工程皮肤奠定了牢固的基础。 AIM:To investigate the possibility of isolation, culture and identification of human epidermal stem cells in vitro to seek for optimal seed cells for the construction of tissue engineered skin. METHODS: The experiment was conducted in the Center for Experimental Animals, College of Basic Medical Sciences from January to December 2002. Based on the rapid adhesion of the epidermal stem cells to type Ⅳ collagen, the stem cells were isolated and purified from the epidermal stem cells which were cultured in the rat type Ⅳ collagen coating culture bottle directly by rapid adhesion, and then underwent the identification of morphology, ultrastructure and immunohistochemistry. RESULTS: The characters of the selected stem cells were in accordance with the characters of epidermal stem cells, indicating that the selected cells were the epidermal stem cells. CONCLUSION: It is feasible to select and culture the epidermal stem cells in vitro aforehand, and such cell colonies are predicted to be the seed cells for the construction of tissue engineered skin. So, it provides a firm basis for constructing tissue-engineered skin taking the epidermal stem cells as the seed cells in the future.
