摘要
目的:研究凉膈散药物血清体外对内毒素刺激小鼠RAW264.7细胞CD14mRNA表达的影响。方法:大鼠以凉膈散灌胃,制备药物血清。以凉膈散药物血清或正常血清的培养液与LPS体外共同培养RAW264.7细胞,原位杂交法检测凉膈散药物血清对内毒素刺激RAW264.7细胞CD14mRNA表达变化。结果:与LPS刺激组比较,不同剂量药物血清组CD14mRNA表达量均比它低,呈剂量依赖性;正常血清组则无显著性差异。结论:凉膈散含药血清体外对LPS刺激小鼠单核细胞CD14mRNA转录具有抑制作用。减少LPS效应细胞的CD14mRNA的表达可能是凉膈散减轻LPS对机体损伤,发挥解毒作用的机制之一。
To study the influence of Lianggesan on the expression of CD14mRNA of macrophage RAW264.7 induced hy LPS . Methotis： The Lianggesan medicated serum was prepared and then the macrophage was incubated with LPS and serum with or without Lianggesan. Collected the specimens and detected the expression change of CD14mRNA by ISH. Results： The express of CD14mRNA of LPS-induce group rises significantly compared with that of the blank-control group, compared with that of LPS-induce group： the different dose groups significantly decrease and it appears dose-dependence, the blank serum group has no significant difference, Conclusion： Lianggesan medicated serum could inhibite the expression of CD14mRNA in RAW264.7 induced by LPS, which may be one of the signal traneduction mechanism of Lianggesan＇s detoxication.
