摘要 目的进一步了解鼠疫反向血球凝集试验的敏感性和特异性。方法用鼠疫EV株和其他试验菌株分别在28℃和37℃培养和EV株感染小白鼠脏器进行鼠疫反向血球凝集试验。结果能检出鼠疫菌EV株2.5×104个菌/m l,与其他菌≥5×108个菌才出现交叉凝集,而且部份液体混浊。EV株37℃培养感染小白鼠72h脏器悬液稀释度1∶2560时能检查出F1抗原。检出率60%~80%。结论鼠疫反向血凝试验敏感性和特异性是好的,鼠疫菌F1在37℃培养才能产生检出、28℃培养不能产生检出。 Objective To study the sensitivity and specificity of the detection of immunoglobulin of Y. pestis EV strain by reverse indirect hem agglutination assay (RIHA). Method Y. pestis EV strain and other strains were cultured at 28℃ and 37℃. Mice were infected with EV strain or other strains cultured at 28℃ or 37℃. The viscera of the infected mice were collected for the detection of the bacteria by RIHA. Results Y. pestis EV strain could be detected at 2.5×10^4 bacteria/ml, while the other bacteria strains could only be detected at 35×10^4 bacteria/ml, showing a turbid solution. F1 antigen could be detected in 1:2560 diluted viscera suspension obtained from the mice at 72h after infection with Y. pestis EV strain (cultured at 37℃), with a positive rate of 60~80%. Conclusion The experimental results demonstrated that the RIHA method for the detection of Y. pestis EV strain has a high sensitivity and specificity.The F1 antigen of Y. pestis EV strain can only be detected in the mice infected with the EV strain cultured at 37℃ butnot those cultured at 28℃.
