摘要
目的研究重组人角质细胞生长因子-2(Recombinanthumankeratinocytegrowthfactor-2,rhKGF-2)涂膜剂对大鼠皮肤伤口愈合速度的影响。方法(1)用预热100℃的100g砝码在大鼠背部脱毛部位烫8s,形成4个直径约2.5cm的深II度烫伤口;(2)手术方法切除大鼠背部部分皮肤,形成2个1.5cm×4cm的皮肤全层缺损伤口。造模后第2天开始每天局部给药1次,连续15d,每4d测量伤口面积并计算愈合率,第16天取伤口及其周围皮肤组织,石蜡切片,Masson’s三色法染色,MOTICAdvanced3.2形态分析软件分析新生上皮面积、厚度和上皮细胞迁移距离。结果rhKGF-2涂膜剂对大鼠皮肤深II度烫伤和皮肤全层切除伤口的愈合均有明显的促进作用,使新生上皮面积、平均厚度和上皮细胞移行距离等明显增加。作用强度比同剂量的单纯的rhKGF-2溶液剂好。结论rhKGF-2涂膜剂能明显加快大鼠皮肤烫伤和皮肤全层切除伤口周围上皮细胞的增生与迁移,加快创面愈合过程。
OBJECTIVE To observe the effect of recombinant human keratinoeyte growth factor-2 (rhKGF-2) liniment on the healing of skin wound. METHODS ( 1 ) The back skin of rat was burned for 8 seconds by 100℃ weights with diameter 2.5cm to become four deep Ⅱ degree burn wounds. (2) Two pieces of full-thickness skin on the back of each SD rat were excised surgically to make two 1.5 × 4cm full-thickness skin loss wounds. One day postinjury, the animals were divided into 6 groups. The wounds were respectively applied topically with saline solution in negative control group, liniment vehicle in vehicle group, rhKGF-2 solution in positive group, and rhKGF-2 liniments (200, 100 and 50 (g/mL) in three experimental groups daily for 15 days. The wound areas were measured at 4^th d, 8^th d, 12^th d and 16^th d. On the 16^th postinjury, skin tissue biopsy specimens of the wound were taken. The area and thickness of neoepithelia tissue and the migrated distances of neoepithelium were detected with MOTIC Advanced 3.2 morphological analysis software after Masson's Trichrome staining. RESULTS rhKGF-2 liniment could accelerate the healing rate of both deep Ⅱ degree burn wound and full-thickness skin loss wound in rats. It could improve reepidermidation and increase the migrated distance, average area and thickness of neoepithelium in a dose-dependently manner. The potency of rhKGF-2 liniment was stronger than that of rh-KGF-2 solution at the same doses. CONCLUSION These results show that rhKGF-2 liniment can significantly accelerate wound healing in rats.
