Expression of transforming growth factor-β and its receptor in human renal cell carcinoma

摘要 目的探讨转化生长因子(TGF)-β及其受体(TGF-βR)的表达与肾细胞癌(RCC)的关系.方法采用逆转录聚合酶链反应(RT-PCR)及表达丰度定量法检测肾细胞癌组织和正常对照组中TGF-β1~3、TGF-βRⅠ~Ⅲ的mRNA表达.结果 RCC组TGF-β1~3表达水平均明显高于正常对照组(P<0.005),而TGF-β1更为显著;RCC组的TGF-βRⅡ表达水平明显低于正常对照组 (P<0.005);而TGF-βRⅠ、Ⅲ的表达水平略高于正常对照组,但差异无显著性(P>0.05).结论 RCC组织中TGF-β的表达较正常肾组织高,而TβRⅡ的表达则明显低于正常肾组织,由此降低癌细胞对TGF-β的敏感性,减弱TGF-β抑制肾癌细胞增殖的作用,对TGF-β、TβRⅡ表达水平的检测有助于RCC的诊断和预后判断;TβRⅠ、Ⅲ的表达水平可能与RCC无明显关系. Objective To appraise the gene expression and relationship of transforming growth factor(TGF)-β and TGF-β receptor in RCC. Methods TGF-β and TGF-βR mRNA were analyzed by RT-PCR in 18 RCC tissues and 18 normal renal tissues (as control) which were collected during operation. Results The TGF-β1~3 gene expression in RCC was higher (P〈0. 005) and the expression of the TGF-βRⅡ was lower (P〈0. 005). But the gene expressions of TGF-βR Ⅰ and TGF-βR Ⅲ were slightly higher than those of normal but were with no significant difference (P〉0.05). Conclusions Expression of TGF-β is increased and that of TβR Ⅱ is decreased in RCC. The decrease of TβR Ⅱ expression may be the cause of impaired sensitivity to TGF-β in RCC suggesting as a significant prognostic factor in patients with RCC. (Shanghai Med J,2005,28:598-600).
