PTEN在脑胶质瘤表达的定性研究

Qualitative Study of PTEN Gene Expression in Human Glioma

目的探讨抑癌基因PTEN在脑胶质瘤的表达情况及其临床意义。方法应用逆转录聚合酶链反应(RT-PCR)方法,检测45例胶质瘤组织与23例正常脑组织PTEN基因外显子1-9、1-8、5-9、5-8 mRNA失表达情况。结果在45例肿瘤组织中,PTENmRNA平均失表达率为55.6%(25/45)。正常脑组织无1例失表达。胶质瘤组织和正常脑组织中PTENmRNA失表达水平存在明显差别(P<0.05)。胶质瘤中PTENmRNA失表达水平与患者年龄、性别无关,但随着恶性程度分级增高其失表达水平明显升高(P<0.05)。结论PTENmRNA表达水平的异常在胶质瘤的发生和发展过程中起着重要作用。

Objective To investigate the expression of tumor suppressor PTEN mRNA in glioma aswell as to elucidate its clinical significance. Methods The expression level of PTENmRNA was detected by reverse transcription polymerase chain reaction in 45 cases of human gliomas and 23cases of normal brain tissues. Results The average negative expression rate of PTENmRNA in human glioma was 55.6% （25/45）. No negative expression was found in normal brain tissues . The negative expression rate of PTENmRNA was increased with progression of pathological grade （P＜0.01）. Conclusion The deletion or lower expression of tumor supressor gene PTEN may play an important role in the development and matastasis of glioma.
