HSV-1载体中介lacZ基因导入大鼠中枢神经系统及其表达的研究

摘要 目的研究用HSV-1载体将外源基因导入大鼠中枢神经系统的方法。方法将HSV-1载体接种于大鼠嗅神经末梢,在不同时间取各部位脑组织检测lacZ基因表达产物β-半乳糖苷酶活性,并观察大鼠脑组织局部及全身的病理学和免疫学变化。结果接种HSV-l载体后2d在嗅球中检测到β-半乳糖苷酶活性,接种后4d在嗅球、嗅前神经核、蓝斑、杏仁核、海马、颞叶皮质、间脑以及最后区均检测到β-半乳糖苷酶活性,并持续10周以上;脑组织及全身各脏器未见病理学及免疫学改变。结论HSV-1载体可通过大鼠嗅神经通路将外源基因导入中枢神经系统并在其中表达。 [Objective] To study the feasibility of transferring foreign gene by HSV-1 vector into central nervous system by olfactory pathway in rats. [Methods] We inoculated HSV-1 vector into peripheral olfactory nerves of rats by nasal instillation and observed β-galactosidase expression in different brain areas and time after inoculation by histo-chemical method. [Results] β-galactosidase was expressed in olfactory bulb after 2 days, and it was expressed in anterior olfactory nucleus, locus ceruleus, amygdaloid nucleas, hippocampus, temporal cortex, diencephalons and area postrema after 4 days. And its expression had lasted for more than ten weeks, No local and systemic pathological and immunological changes were seen in the treated rats. [Conclusions] The HSV-1 vector could be used to transfer foreign gene into central nervous system by olfactory pathway.
