摘要
目的了解E-选择素(E-selectin)、整合素β(1Integrinβ1)、免疫球蛋白超家族成员(ICAM-1)在人胃癌细胞中的表达水平,探讨这3种细胞粘附分子与胃癌的关系。方法采用NothernBlotting法和ELISA法检测胃癌细胞、正常胃上皮细胞和人脐静脉内皮细胞上E-selectin、Integrinβ1及ICAM-1的表达水平并进行比较。结果胃癌细胞、正常胃上皮细胞和血管内皮细胞均有E-selectin、Integrinβ1及ICAM-1的表达。但胃癌细胞3种粘附分子表达水平均高于正常胃上皮细胞和人脐静脉内皮细胞,差别均有显著性意义(P<0.05)。结论E-selectin、Integrinβ1、ICAM-1可能与胃癌细胞转移有关。
Objective To understand expression of the E-selectin,lntegrin β1. and ICAM-1 in gastric cancer cell line, investigate the relationship between cell adhesion molecules and gastric cancer cells. Method Expressions gene in three cell lines were studied by Northern blot hybridization. The supernant concentrations of the E-selectin,Integrinβ1 and ICAM-1 were measured with ELISA in three cell lines. Result mRNA eof the E-selectin,Integrinβ1 and ICAM-1 all expressed in three cell lines. The expression level of three adhesive molecules in gastric cancer cell line detected by ELISA were higher than that of normal stomach epithelium and umbilical vein endothelial cells, and there was significantly difference between them re-spectively (P〈0.05). Conclusion The there adhesive molecules (E-selectin,Integrin β1,ICAM-1)may be related to the tumor metastasis.
