摘要
用抗牛精子膜伴刀豆素A结合糖蛋白血清,在兔、小鼠、地鼠和人精子表面进行了免疫细胞化学定位观察。结果表明,几种动物和人的精子均被特异性标记。标记的主要部位是顶体区,与伴刀豆素A亲和细胞化学的标记相似。地鼠精子用抗血清封闭后,进行精子-卵子透明带的体外结合试验,证明精子与卵子透明带的结合受到显著抑制。本实验结果表明,精子膜伴刀豆素A结合糖蛋白抗原具有种间交叉反应性,并且参与精子与卵子透明带的识别过程。
The antigenic property and the role in sperm-ovum recognition of the concanavalin A-binding glycoprotein on the sperm plasma membrane were studied by PAP immunocytochemical method and sperm-zona pellucida binding assay with the antiserum against to the glycoprotein seperated from plasma membrane of bovine sperm.In immunocytochemical study,all the sperms of rabbit,mouse,hamster and human showed positive staining,mainly at the acrosomal region,The staining pattern was similar to the affinity cytochemical staining of HRP labeled concanavalin A. In the sperm-zona pellucida binding assay,the number of sperms bound to ovum zona pellucida was significantly reduced when the sperms were treated with the antiserum to concanavalin A-binding glycoprotein. The present study indicates that the concanavalin A-binding glycoprotein of sperm plasma membrane has the antigenic property of species-cross reactivity and may be implicated in sperm-zona pellucida recognition.
