大鼠抗小鼠胸腺基质细胞单克隆抗体的鉴定 被引量:2

CHARACTERIZATION OF MONOCLONAL ANTIBODIES PRODUCED BY RAT SPECIFIC TO MURINE THYMUS STROMAL CELLS

摘要 根据小鼠胸腺冰冻切片免疫组织化学染色,将已获得的30种大鼠抗小鼠胸腺基质细胞单克隆抗体分为8组:1.髓质胸腺基质细胞(TSC)阳性,皮质TSC阴性;12.髓质部分TSC阳性,皮质TSC阴性;3.皮、髓交界部分TSC阳性;4.皮、髓质部分TSC阳性;5.髓质TSC阳性,皮质部分TSC阳性,6.皮、髓质大部分TSC阳性;7.被膜和血管内皮细胞呈阳性;8.髓质TSC和皮质胸腺细胞为阳性。用这8组单抗对本室建立的5株小鼠胸腺基质细胞系进行免疫荧光染色,发现它们与各细胞系的反应各不相同。对这些单抗的鉴定不仅有利于鉴别TSC亚群,并且为研究T细胞与基质细胞的相互作用提供了新的抗体。 By means of intrasplenic immunization,two series of rat anti-MTSC (mouse thymic stromal cells, MTSC) hybridoma have been established and 30 cell lines which secrete McAbs have been obtained. The supernatents from cell lines secreting McAbs have been identified by immunohistochemical staining of mouse thymic frozen section : (1) react only to medullary TSC, not to cortical TSC; (2) react only to a subpopulation of medullary TSC; (3) react only to a subpopulation of cortex/medulla junction TSC; (4) react to a subpopulation of cortical and medullary TSC; (5) react to majority of medullary TSC and a subpopulation of cortical TSC; (6) react to majority of medullary and cortical TSC; (7)react only to epithelial cells of capsular and endothelial cells of vessels; (8)react to medullary TSC and thymocytes of cortex. It is observed that 8 groups McAbs were reacted with different MTSC lines by indirect immunofluorescence staining,and their reactions to various MTSC lines are different. Our results are not only useful for identification of TSC subsets and also important for investigation of T cell development.
