杧果炭疽菌随机扩增多态性DNA分析 被引量:6

摘要 对海南、广东、四川、广西等14个杧果主要栽培地采集的70个杧果炭疽菌分离菌株进行了遗传多态性的初步研究。利用19个10碱基随机引物对70个杧果炭疽菌分离菌株基因组DNA进行RAPD分析。根据它们的遗传相似性,这70个杧果炭疽菌分离菌株被聚类为2大群(I和II)和相似系数为0.306 ̄0.928的6小群(I-a,I-b,I-c,I-d,II-a,II-b)。不同栽培地的相似系数低;同一栽培地的相似系数高,所以杧果炭疽菌的遗传基础与地理来源虽没有严格的对应关系,但存在相关性。抗药性试验结合RAPD分析结果表明,抗药性菌株一般与非抗药性菌株的亲缘关系较远,推测遗传基础的改变是抗药性产生的原因之一。 A preliminary study was carried out on their genetic diversity of 70 mango anthracnose isolates from 14 major planting areas in the provinces of Hainan, Guangdong, Guangxi, Sichuan, etc. The genetic polymorphisms of their genomic DNA were studied by RAPD with 19 pairs of 10-bp arbitrary primers and were clustered on coefficients. The 70 isolates were clustered into 2 groups (Ⅰ and Ⅱ) and 6 subgroups (Ⅰ-a, Ⅰ-b, Ⅰ-c, Ⅰ-d, Ⅱ-a and Ⅱ-b) based on their genetic similarities, and these subgroups had a similarity coefficient of between 0.306 and 0.928. The isolates from different locations were clustered on low coefficients and those from the same location clustered on high coefficients. This indicated that the genetic basis of mango anthracnose was correlated, although not strictly, with its geographic source, and that the genetic background of the mango anthracnose was similar in the same location.
