摘要
目的建立大鼠非酒精性脂肪性肝炎(NASH)的动物模型。方法雄性SD大鼠20只,随机分为模型组(n=10)和正常对照组(n=10)。模型组喂养高脂饮食,即在普通饲料基础上加10%猪油、2%胆固醇、5%蛋黄粉。正常对照组喂养普通饲料。喂养12周后,观察-般情况,检测大鼠血清生化指标及炎症介质,并行肝组织病理检查。结果模型组体重、肝体比、血清甘油三脂(TG)、肿瘤坏死因子α(TNFα)均高于正常组;病理学均出现弥漫性肝细胞脂肪变性,且90%存在小叶内炎症,80%出现汇管区炎症;电镜示肝细胞索排列紊乱,胞浆脂滴大量出现、聚积、线粒体嵴肿胀,肝星形细胞和枯否细胞增生,细胞核变形,部分呈现核固缩表现。结论通过12周的高脂肪、高胆固醇饮食成功地制备出大鼠NASH模型。
To establish a nonalcoholic steatohepatitis(NASH) experimental model of rats. Methods 20 male SD rats were randomized to two groups: one group fed with high fatty diet consisting of standard diet, 10 % lard, 2 % cholesterol, 1% egg yolk,and the other group fed with standard diet as control. The pathological structure change of rat's liver cell was observed and serum lipid value and inflammatory factor were detected 12 weeks laten. Results The experimental group was higher than control group in weight, ratio of liver weight to body weight, triglycerides and TNF-a. There was significant hepatocellular steatosis in pathology, some of which existed inflammation in lobular and portal part. The incidence of NASH in rats with ligh fatty diet was 100 percent. Conclusion Through 12 weeks fatty foods feeding, and effective experimental rats model of NASH is established successfully.
