摘要
目的探讨PD大鼠移植后神经元凋亡的时程特点及褪黑素的抗凋亡作用.方法将大鼠PD模型分为3组,即胚鼠腹侧中脑组织(VM)移植组、褪黑素(MT)干预组及对照组;采用原位末端标记法(TUNEL)结合流式细胞术及免疫组化方法,对3组的神经元凋亡情况进行定量分析.结果神经细胞在移植后1~5天凋亡达到高峰,此后逐渐平缓下降;褪黑素干预组各个时相点凋亡细胞数均明显低于胚VM移植组.结论 PD大鼠移植后1~5天是细胞凋亡的高峰期;褪黑素具有明显的抗凋亡作用.
AIM To explore the phase of grafted embryonic dopamine neurons apoptosis and the regulation by melatonin(MT) and provide experimental basis for clinical therapy. METHODS Terminal deoxynucletidyltransferase-mediated biofinylateddeoxyuyidinetriphosphate nickendlabeling technique combined with fluid cytometre was used to quantiatively detect apoptotic cells in grafted E14 Rat Venrtal Mesencphala group; MT treated group and control group. RESULTS The nember of apoptotic cells reached the peak at 1 d to 5 d after neural transplantation, then declined. But the nember of apoptotic cells in MT treated group was significantly decreased at each observation points. CONCLUTION The Time Course of apoptosis of grafted embryonic dopamin neurons peaked at 1 d to 5 d, which could be attenuated throuth MT.
