姜黄素对单个核细胞组织因子表达的影响 被引量:1

摘要 目的观察姜黄素(curcumin,CUR)对内毒素(lipopolysaccharide,LPS)诱导外周血单个核细胞(MNC)组织因子表达的影响。方法人外周血MNC的分离采用淋巴细胞分离液,MTT法检测细胞活性,细胞促凝活性的检测采用一期凝固法。结果 LPS可以剂量依赖性地诱导MNC组织因子的表达(20.1±5.1mU/10~6 cells vs 7.3±2.3 mU/10~6cells,P<0.01),CUR可以显著抑制LPS诱导的细胞冻融液组织因子活性(14.6±3.6 mu/10~6 cells vs 24.5±6.4 mU/10~6cells,P<0.01)。且CUR提前60 min加入MNC,其抑制作用最强。AngⅡ(10^(-7)M)可加强LPS诱导MNC细胞冻融液TF活性(50.7±12.3 mU/10~6ceHs,vs 24.1±7.2 mU/10~6cells,P<0.01),CUR对该作用也有显著的抑制作用。结论姜黄素可显著抑制LPS以及Ang Ⅱ+LPS诱导的MNC组织因子的表达。 Objective To study the cffects of curcumin (CUR)on the tissue factor expression of peripheral blood mononuclear cells (MNC). Methods MNC were isolated from herlthy volunteers by lymphoprep sedimentation. MTT assay was used to evaluate cytotoxicity of CUR. Precoagulant activity (PCA) was determined with one stage clotting method. Results The tissue factor activity expression of MNC increased significantly in the lipopolysaccharide(LPS) group(20. 1 ± 5. 1 mU/10^6 cells vs 7.3 ±2.3 mU/10^6cells,P 〈0.01 ). MNC were preincubated for 1 h with different concentrations of CUR( 1,4,8 μM) followed by treatment with LPS(4 μg/ml) for 6 h, the results showed that CUR inhibited the induction of the TF activity in a dose dependent manner. The results of time courses for CUR to inhibit LPS induced TF activity in MNC showed that the inhibitory activily of CUR was the most powerful when the cells were treated with CUR for 60 min before the addition of LPS to the MNC. Ang Ⅱ (10^-7M)upregulated tissue factor expression of human MNC induced LPS, and significantly inhibited these effects. Condusion CUR may significantly inhibit the expression of tissue factor in MNC induced by LPS and Ang Ⅱ + LPS.
