薄荷醇对大鼠全脑缺血-再灌注模型脑组织凋亡基因的表达 被引量:3

The Expression of Menthol on Apoptosis Gene(c-fos,bcl-2) of Brain Tissue of Cerebral Inschemia-reperfusion Injury Model of Rates

摘要 目的观察薄荷醇在大鼠脑缺血再灌注损伤时对脑组织凋亡基因(c fos,bcl2)的表达的影响,进而来研究薄荷醇对脑有保护作用。方法用pulsinelli Brierleyr四血管闭塞的方法制作急性全脑缺血再灌注损伤的模型。将大鼠分为正常组、缺血再灌注对照组、缺血再灌注薄荷醇组,后者又根据注射薄荷醇不同时间点分为C1(5min);C2(15min);C3(30min);C4(45min);C5(60min)。处死大鼠,采用免疫组化的方法检测脑组织中的凋亡基因的表达情况。结果缺血再灌注后皮质、海马等脑区均有c fos表达,缺血再灌注表达增强,缺血再灌注薄荷醇组则明显抑制c fos表达。bcl2表达结果与c fos相反。结论薄荷醇对脑有保护作用,其作用可能与下调c fos,上调bcl2表达有关。 Objective To study the expression of menthol on apoptosis gene (c-fos, bcl-2) of brain tissue of cerebral inschemia-reper{usion injury model o{ rats, so as to study the protection of menthol to brain. Methods The acute cerebral inschemia-reper{usion injury model was made with Pulsinelli-Brierley four -vessel occlusion method. Dividing the rats into the normal group (A), the inschemia-reper{usion control group (B), and the inschemia-reperfusion with menthol injecting group (C), which was divided into five subgroups based on different time injecting menthol (C1 5min, C2 15min, C3 30min, C4 45min, C5 60min). After executing the rats, analysing the expression on apoptosis gene o{ brain tissue by the immunohistochemical method. Results The expression of c-fos was detected in the cortex and hippocampus area. It was rein{orced in group B, while inhibited in group C. The expression of bcl-2 was just the opposite of c-fos. Conclusion Menthol can protect brain. Its mechanism may be related to the expression of lowering c-fos and raising bcl-2.
