Determination of triptolide in tripterygium rough- preparation by HPLC

目的:建立高效液相色谱法测定雷公藤粗制品中雷公藤甲素的含量.方法:雷公藤粗制品经洗脱提取后,采用十八烷基键合硅胶柱,以乙腈-水(30:70)为流动相,以218 nm为检测波长,检测雷公藤粗制品中雷公藤甲素的含量.结果:雷公藤甲素在1.92～11.52μg/ml范围内线性良好,r=0.999 9,平均回收率为95.32%,RSD为1.16%.结论:本法操作简便、结果准确,可用于雷公藤粗制品中雷公藤甲素的含量测定.

Object： To determine the content of triptolide in tripterygium rough-preparation from Leigongteng （Tripterygium Wilfordii Hook. F. ）. Methods ： The rough- preparation was eluted with acetonitrile on a neutral alumina cartridge. Then, the content of triptolide was measured by HPLC on a C18 column using acetonitrile-water （30：70，v/v） as mobile phase. The UV detection was set at 218nm. Results： The method proved to be linear in the range of 1.92-11.25μg/ml with a regression coefficient of 0. 999 9. The average recovery of triptolide was 95.32% and the RSD was 1.16%. Conclusions：This method was rapid and accurate for the measurement of triptolide in tripterygium rough- preparation。
