微波固相合成寡糖工艺的初步研究

摘要 以葡萄糖(C6H12O6.H2O)为原料,以杂多酸为催化剂对生物活性物质寡糖的微波固相合成进行了研究。确定最佳反应条件为:微波功率800瓦,微波辐射时间10min,引发剂水的添加量30%,催化剂酸添加量10%;产物得率为50.6%。经高压液相色谱分析产物,确定微波固相合成物的组成为:葡萄糖49.34%,麦芽糖10.03%,异麦芽糖18.03%,麦芽三糖5.40%,潘糖10.13%,异麦芽三糖5.30%,四糖和五糖占1.61%。 The synthesis of bioactive oligosaccharide with glucose and phosphoric acid as the catalyst by solid state reaction under microwave irradiations was studied. The optimum reaction conditions was found as follows: microwave power 800w ,microwave irradiation time 10 rain, adding 30% water to initiate reaction, adding 10% heteropoly acid A as the catalyst. The yield of product was 50.6%. The product was characterized by HPLC analysis. The constitution of product was established as follows: glucose 49.34%,maltose 10.03%,isomaltose 18.03%, maltotriose5.40%, panose 10.13% , isomaltotriose5.30%, tetrsaccharides and pentasaccharides 1.61%.
