摘要
目的观察流体剪切应力对人骨髓间充质干细胞增值及细胞周期的影响。方法人骨髓间充质干细胞受强度为3 dyne/cm2的流体剪切力刺激30 m in,应用四甲基偶氮唑蓝(MTT)法和流式细胞仪检测细胞的增殖能力和细胞周期。结果流体剪切力作用后,细胞增殖能力提高,细胞活性增强。S期细胞百分比(13.53±1.47)%较对照组细胞的S期百分比(7.45±2.65)%约增高近181.6%。结论流体剪切力对人骨髓间充质干细胞具有提高细胞增殖能力及增强细胞活性的作用。
Objective To observe the effect of fluid shear stress (FSS) on the cellular proliferation and cell cycle of the human bone marrow mesenchymal stem cells (hMSCs). Methods After hMSCs were stimulated by the FSS (3 dyne/cm^2, 30 min), MTT assay and flow cytometry were applied to measure the cellular proliferation and cell cycle. Results FSS treatment accelerated cellular proliferation obviously. After 24 h from the end of FSS stimulation for 30 min, the percentage of phase S cells [ ( 13.53 ±1.47 ) % ] was increased to 181.6% as compared to that of the control group [ (7.45± 2. 65) % ]. Conclusion FSS has the enhancing effect on the cellular proliferation and cell cycle of hMSCs.
