p185高表达人卵巢癌裸鼠模型的建立

Establishment of nude mice model of human ovarian carcinoma exhibiting high p185 expression

目的建立高表达p185的人卵巢癌裸鼠模型。方法BALB/c裸鼠皮下接种高表达p185的人卵巢癌细胞株SKOV3,观察成瘤情况及移植瘤病理组织学特点,并用SP免疫组织化学法测定移植瘤的p185表达情况。结果12只裸鼠均荷瘤成功,病理组织学与免疫组化均证实移植瘤保持了原细胞系特征。结论高表达p185人卵巢癌裸鼠模型的成功建立为抗p185抗体药物的体内药效研究提供了理想的平台。

Objective To establish an animal model of human ovarian cancer overexpressing p185 in nude mice. Methods A human itvarian cancer cell line SKOV3 was administered suhcutaneously to trade mice, The tumor growth and histopathological features were observed, and p185 level in tumor cell surface was evaluated by SP immunohistochemistry. Results Twelve nude mice were successfully transplanted with the tumor. The features of the transplanted tumors were the same as the primary cell line in histopathological and immunohistochemical assays. Conclusion The animal model of human ovarian cancer overexpressing p185 is established successfully. It provides a useful tool for studying the effect of anti-p185 antibodies on tumor growth.
