摘要 目的:观察新型发光纳米材料—无机纳米晶体颗粒量子点对小鼠骨髓造血细胞的标记和成像,探讨它在血液细胞分化发育研究中的作用。方法:采用最大发射波长为550nm的绿色量子点转铁蛋白复合物分别在体外和体内对小鼠骨髓造血细胞进行标记研究,并观察量子点转铁蛋白复合物对骨髓造血干细胞CFU-GM的影响。结果:量子点转铁蛋白复合物可标记骨髓中大部分造血细胞,而且荧光强度强,光学和化学稳定性好,对骨髓造血干细胞的增殖分化功能无影响。结论:量子点在血液细胞成像方面表现了优良的特性,在细胞生物学研究中具有重要作用。 Objective: To observe the labeling and imaging of mouse bone marrow hematopoietic cells with the new luminescent inorganic nanocrystals-quantum dots and study the effect of quantum dots on the proliferation and differentiation of hematopoietic cells. Methods: The bioconjugate of green quantum dots with mouse transferrin was used to label the mouse bone marrow hematopoitic cells both in vitro and in vivo, the influence of the bioconjugate to the CFU-GM was also observed. Results: The quantum dots bioconjugate could label most mouse bone marrow hematopoietic cells with strong fluorescence intensity and good optical and chemical stability and no influence to the proliferating and differentiating function of hematopoitic progenitors at the same time. Conclusion: Quantum dots exhibit extremely desirable properties for cell imaging. In the future, quantum dots will play a vital role in the study of cell biology.
