两种血清苯丙氨酸高效液相色谱测定法的比较 被引量:6

摘要 目的对两种血清苯丙氨酸高效液相色谱测定法进行比较.方法对高效液相色谱-紫外检测法(HPLC-UV)和高效液相色谱-荧光检测法(HPLC-FLD),从线性范围,检测限,精密度等参数上进行比较,用两种方法分别测定了正常儿童和一组PKU患儿血清phe值,对结果进行统计分析.结果HPLCUV法和HPLC-FLD法的线性范围分别为6~1512 μmol/L和12~1220 μmol/L,最低检测限分别为0.75 μmol/L和3μmol/L,平均回收率分别为98.6%和102.5%,批内CV均<5%,批间CV分别为4.01%和7.17%,正常儿童血清phe值分别为63.2±11.6 μmol/L和92.8±21.3μmol/L(x眘);两种方法同时测定PKU血清phe结果之间的差异有显著性的意义(P=0.019),HPLC-FLD法值较高但与HPLC-UV法值呈正相关(r=0.783).结论两种HPLC方法均能灵敏,特异地测定血清phe,满足对pku筛查的需要,HPLC-UV法在精密度及缩短分析检测时间上略优于HPLC-FLD法. Objective Comparison the two high performance liquid ehromatography(HPLC) for serum phenylalanine(phe). Methods HPLC-ultraviolet absorbing detection(HPLC-UV) and HPLC-fluorescence deteetion(HPLC-FLD) were compared in linear range, recovery rate detection limit and precision. Identical samples were detected with two HPLC methods, The results were analyzed. Results The linear range, mean recovery rate, in-run CV and between-run CV were 6 - 1512 μmol/L, 0.75 μmol/L, 3.68% and 4.01% for HPLC-UV, respectively, and 12 - 1220 μmol/L, 3/maol/L, 102.5%, 4.07% and 7.17% for HPLC-FLD, respectively. Conclusion Both HPLC are sensitive, accurate, simple, rapid and adapted for clinical use. The HPLC-UV is more presision and rapid than the HPLC-FLD.
