HPLC-ELSD法测定黄芪甲苷的实验研究

Determination of content of astragalin from oral compound astragalus soup by using HPLC-ELSD

目的研究复方黄芪口服液中黄芪甲苷的HPLC-ELSD测定方法。方法采用蒸发光散射检测器(ELSD)对复方黄芪口服液中的黄芪甲苷进行HPLC分析。色谱柱:AlltechC18柱(5μm,250×4.6mm);流动相:乙腈-水(36:64);流速:1.0ml/min;漂移管的温度为110℃,雾化气流速为2.3L/min。结果此方法线性良好,线性范围为0.997～15.95μg,R=0.9994;平均加样回收率为95.4%,RSD为4.2%。结论此方法简便快捷,精密度、准确性、重现性均能满足质量控制的要求,是一种比较理想的复方黄芪口服液中黄芪甲苷含量的测定方法。

Obiective To develop a method to determine Astragaloside Ⅳ in compound Radix Astragali oral solution by HPLC-ELSD. Methods HPLC-ELSD was used for analyzing Astragaloside Ⅳ in compound Radix Astragali oral solution, with All-tech C18 coluran（5μm, 250×4.6mm） ,and acetonitrile-water（36：64）as a mobile phase. The flow rate was 1.0ml/min. The temperature of drift tube for ELSD was 110℃ and the flow rate of nebulizing gas was 2.3L/min. Results The calibration curve was liner in the range of 0.997～15.95μg , R=0.9994. The recvery of the added sample was 95.4% and RSD was 4.2%. Conclusion The method is simple, rapid, more precision, accurate and reproducible, and it provides an effective way for quality control of Astragaloside Ⅳ in compound Radix Astragali oral solution.
