摘要
通过响应面分析(RSA)法,优化了姬松茸深层发酵菌丝体胞内多糖的热水浸提条件,即提取温度为95℃,时间为3·5h,料(湿菌体)水比为1∶3·5(w/w)。结合得率及对体外静止脾淋巴细胞增殖的促进实验结果,确定了菌丝体粗多糖的最适醇沉浓度为85%。在上述工艺条件下实际测得姬松茸深层发酵菌丝体粗多糖的提取得率为77·26g/kg,所得粗多糖在20mg/(kg·d)的剂量下对小鼠的S180移植瘤的抑瘤率为59·84%。
Response surface analysis was applied in hot water extraction of polysaccharide from mycelia of A. blazei Murill. The technological condition parameters were determined using regression analysis. The optimum conditions were as follows: extraction temperature of 95℃ , extraction-duration of 3.5h, water to material(fresh mycelia) ratio at 3.5:1(w/w). Final ethanol concentration of 85 % was determined by the experimental results of both yield and proliferation of no-active spleen lymphocyte 1 in vitro. Under the above conditions, the yield of crude polysaccharide from mycelia of A. blazei Murill reached 77.26 g/kg and the inhibition rate aganist S180 in mice in vivo was 59.84%.
