摘要
目的研究血链菌生物膜形成中死菌/活菌的空间分布。方法在模拟人口腔环境的简易人工口腔模型内,形成血链菌生物膜,应用激光共聚焦扫描显微镜和死菌/活菌荧光染色方法,对血链菌生物膜形成中死菌/活菌的空间分布进行观察。结果生物膜底部的活菌百分比为20%;随着生物膜厚度的增加,活菌百分比达到70%,其间存在许多黑色气泡样的间隙;随后活菌百分比逐渐减少,至生物膜顶部时几乎全部为死菌。结论生物膜的底部和顶部主要由死细菌组成,而中间层主要是由围绕在黑色气泡样孔和通道周围的活细菌组成。
Objective To examine the spatial distribution of dead/vital cells in the Streptococcus sanguis biofilm. Methods Streptococcus sanguis biofilm was developed in the artificial mouth. Confocal laser scanning microscopy and dead/vital fluorescent stainning technique were combined to study the distribution of dead/vital cells. Results The percent of vital cells was 20% at the bottom of the biofilm, with biofilm depth increased, approached to 70% , and then gradually decreased. Conclusion The bacterial vitality increased from the bottom to the central part of the biofilm and then decreased in the outer parts of the biofilm; many voids were outlined by layers of vital bacteria.
