硫代巴比妥酸测定脂质过氧化物方法的改进

IMPKOVEMENT OF MEASUREMENT OF LIPID PEROXIDE BY THIOBARBITURIC

为提高硫代巴比妥酸（ＴＢＡ）比色法测定红细胞过氧化物的灵敏度，对该法的实验过程进行了两项改进：用５０ｍｍｏｌ／Ｌ的亚硫酸钠代替水作为溶血剂；用２０％三氯乙酸代替冰醋酸作反应的酸性介质。实验结果表明吸光度可提高２．８倍，在２．５～５０ｕｍｏｌ／Ｌ内线性良好，日内重复性实验ｘ±ｓ、ＣＶ分别为２１．７８±０．６９ｎｍｏｌ／ｇＨｂ和３．１７％，日间重复性实验ｘ±ｓ、ＣＶ分别为２０．１７±０．５２ｎｍｏｌ／ｇＨｂ和２．５８％，低、高两管浓度的平均回收率为９８．２５％和１０１％。６０例正常参考值２７．１７±４．

The laboratory method has been reformed for the purpose of improvingthe sensitivity of measurement of erythrocyte peroxide with thiobarbituric acid by colorimet-ric method. 50 mmol/L sodium suifite,instead of water,was used for hemolytic agent and20% trichloroacetic acid , instead of glancial acetic acid, was the acid medium, The resultshowed a good filiform within 25～50 umol/L and 2.8 times increase in absorbance, The val- ue of x±s and CV obtained by repeated test within one day were 21. 78±0. 69 nmol/g Hband 3.17%respectively, and within daytime were 20. 17±0. 25 nmoL/g Hb and 2. 58%,Theaverage rate of recovery for low and high tubes density were repectively 98.25%and 101%. The normal reference value of the sixty cases was 27. 71±4.32 nmol/g Hb.
