摘要
应用聚合酶链反应(PCR)放大口腔癌组织中H-ras癌基因部分DNA顺序,再以特异突变的寡脱氧核苷酸(ODN)探针杂交方法检测口腔癌中H-ras癌基因第12位密码子的G→T突变。结果在21例口腔癌中有6例存在这种突变(28.5%),而10例正常门腔粘膜组织中无1例发生突变。提示:H-ras癌基因第12位密码子→T突变可能是口腔癌中癌基因激活的方式之一。
he polymeras chain reaction(PCR) was used to amplify the DNA sequence of
the Ha- rasoncogene in the tissue of oral cancer, and the G-T mutation at code 12 of the Ha-
rasoncogene was detected by hybridization with mutation specific oligondeoxyucleotide probes
.The result showed that the G-T mutation rate was 28.5%( 6/21) in human oral cancers, butabsent
in 10 normal oral mucosal tissues.This result implicates that G- T mutation atcodon 12 of the
Ha-ras oncogene may be one of the oncogene activated mechanisn in oralcancers .
