摘要
以大麦花粉胚胎发生高频和低频品系Igri和Harrington为材料,采用SDS-PAGE法研究了低温预处理和甘露醇预处理过程中花药可溶性蛋白质的变化。结果发现:Igri花药经过低温预处理后能产生一种低分子量的蛋白质,不同天数之间无差异。而在第14d处产生2种特异的蛋白质。Harrington经过低温预处理后只能引起花药蛋白质含量下降,没有产生特异蛋白质。2种基因型材料经过甘露醇预处理后均能引起花药蛋白质快速降解。但是Harrington从预处理一开始即产生一种特异蛋白质,一直保持到第7d,含量变化不大;而Igri经过甘露醇预处理以后,没有产生任何特异蛋白质。
Water-soluble proteins, which were extracted from anthers of the high-androgenesis line(Igri) and the low-androgenesis line(Harrington) at different stages. of the cold pretreatment and the mannitol pretreatment, were analyzed under denaturing conditions in one-dimension by polyacrylamide gel electrophoresis. The protein patterns revealed qualitative and quantitative differences in size. First, for Igri, the cold pretreatment caused the anthers to accumulate a lower molecular weight protein. There were no differences between the different stages. For Harrington, there were no qualitative differences except the quantitstive differences in the protein pattern. The cold pretreatment only caused the proteins of anther to hydrolyze. Second, the proteins of anther of two genotypes which were treated by 0.3 mol / L mannitol solution were hydrolyzed quickly. Although there was no special protein showed for Igri, a basic protein was detected during the pretreatment period for Harrington.
