摘要
粉防己甲素(Tet)可逆转耐药细胞株K_(562)/VCR对长春新碱(VCR)、足叶乙甙(VP—16)、阿霉素(Dox)和柔红霉素(Dau)的多药耐药。2×10^(-7)~5×10^(-6)μmol/L处理细胞后,VCR、VP—16、Dox和Dau对K_(562)/VCR的半数抑制剂量(IC_(50))分别减少3.4~45.5、1.5~38.5、2.6~9.7和3.2~33.1倍。1×10^(-6)μmol/L Tet与0.02~0.2μmol/L的上述抗癌药共同处理细胞后,其细胞集落数分别减少44.3~84.5、56.4~79.0,52.6~97.4和63.9~10.0%。本文结果表明,Tet是一有效的肿瘤多药耐药逆转剂。
The effects of Chinese medicine, tetrandrine (Tet) on reversal of multidrug resistance of drug-resistance leukemia K562/VCR cells were studied. Cytotoxicity and colony formation were used as assaymethods. The results showed that Tet significantly potentiate the cytotoxicities of VCR, VP-16, Dox and Dau in drug -resistance leukemia K562/VCR cells. After exposure of the cells to 2×10-7~5×10-6μmol/L Tet for 72h, the IC50 values of the four drugs decreased by 3. 4~45. 5,1. 5~38.5,2.6~9. 7 and 3.2~33.1 folds respectively. The cells were culture with 1×10-6μmol/L Tet and 0. 02~0.2μmol/L of the four drugs, the colony numbers of groups of VCR, VP-16, Dox and Dau decreased by 44. 3~84. 5,56.4~79.0.52.6~97.4 and 63. 9~100% respectively. These results showed that Tet was an effective reversal agent of multidrug resistance in tumor cells.
