内耳扫描电镜标本制备技术中ODO法与苯乙烯包埋割断法的比较

摘要 报告应用锇酸-二甲基亚砜-锇酸冷冻割断法(简称ODO法)与苯乙烯树脂包埋组织割断法制备豚鼠内耳扫描电镜样品,并进行了观察比较。结果显示,苯乙烯包埋割断法比ODO法操作更为简便;样品剖面光滑,能够清晰显示内耳组织细胞的内部结构和血管周围间隙,特别适用于微小样品的定向割断。表明苯乙烯包埋割断法是一种较为理想的制备内耳样品的方法。 Abstract O5O4-DMSO-O5O4(ODO) method was compared with styrene embedding cracking method for scanning electron microscopy(SEM). The results showed that the cross section of the samples cracked by resin embedding method appears to be quite smooth and exhibits much clearly the fine structure of the tissues and cells and the perivascular spaces. Furthermore, the preparative process in resin embedding cracking method is simpler, and its results are much more steady than those of ODO method. The styrene embedding cracking method is, especially, adapted to directional cracking of the fine samples of inner ears. Therefore, it is an ideal method for studying the ultrastructure of inner ears.
