摘要
本文介绍了用磷酸甘油酸变位酶测定2,3-二磷酸甘油酸的比色法。探讨了该法的最适反应条件,最适pH7.5~7.6。线性范围可达6.0mmol/L。三份不同浓度标本批内CV1.1%~2.1%,批间CV2.5%~3.9%。60例南京地区健康成人2.3-二磷酸甘油酸含量为1.6~2.7mmol/L,平均回收率94%,与Sigma试剂盒比较测定结果无显著性差异(P>0.05),相关性良好,r=0.982。
In this paper.We reported a colorimetric determination for blood 2,3-Diphosphoglyceric Acid(2,3-DPG) by phosphoglycerate mutase(PGM),We studied optimum conditions of this method and optimum pH was 7.5~7.6.The liner range was up to 6.0 mmol/L. The coefficient of variation of three samples in vary concentration was 1. 1%~2. 1% intr-assay and 2.5%~3.9% inter-assay.The reference value in blood 2, 3-DPG of 60normal adults in Nanjing area was 1. 6~2. 7 mmol/L. The mean recovery rate was 94%. The results estimated by our kits correlated closely with those obtained by Sigma kits and the correlation coefficients was r=0. 982. This method is rapid, simple and accurate. The regent had a long term stability and was easy to buy and was convenient In use.
