油菜花粉变应原的分离纯化及变应原性实验研究 被引量:4

摘要 油菜花粉经硫酸铵沉淀,SephadexG-25凝胶过滤,分成组份A和组份B,前者有明显变应原性。组分A经DEAE纤维素层析,低盐浓度洗脱得组份C,高盐浓度洗脱得组份D。前者为主要变应原,100μg/ml触发肥大细胞,组胺释放率为55.0%。油菜花粉主要变应原组份C是一碱性糖蛋白,分子量为35715.6u,含糖串11.4%,N-末端为组氨酸。等电聚焦电泳分折,有两种不同等电点区带,为pI7.7和7.1。 The allergenicity of Brassica campestris pollen components was studied.The purified allergen,component C,was obtained from this pollen by ammonium sulfate precipitation,Sephadex G-25 gel andDEAE cellulose chromatography. The component C is a basic glycoprotein containing 11.4% of carbohydrate. It’s molecular weight is 36715.6u with histidine at N-terminal.Two subfractions with pI 7.7 and pI7.1 wereseparated by ixoelectrofocuxing electrophoresis. It was reflected that the component C had the charge hetro-geneity.The allerglc rat mast cells histamine release rate was of 55.0%as triggering with 100μg ml of com-ponent C. The component C is the main allergen of Brassica campestris pollen.
