巢式聚合酶链反应检测脑脊液中单纯疱疹病毒的DNA

摘要 应用巢式(nested)聚合酶链反应(PCR)检测患者脑脊液中单纯疱疹病毒(HSV)DNA,25份经鞘内lgG抗体确诊的“单纯疱疹病毒性脑炎(HSE)”患者的CSF标本中23份PCR阳性;105份lgG阴性的“散发性脑炎”患者CSF中15份PCR阳性,其中7份阳性标本为发病1周内所取,2份取于发病当日.提示PCR更适于HSE的早期诊断. Polymerase chain reaction (PCR) assay with two nested primer pairs was developed for the amplification of Herpes simplex virus (HSV) DNA in 25 cerebrospinal fluid (CSF) samples from consecutive patients with Herpes simplex encephalitis (HSE) verified by the finding of intrathecal production of IgG antibody to the virus. PCR detected HSV DNA in 23 CSF samples. While among 105 CSF samples from patients with sporadic en-cephalitis which were negative by IgG, 15 samples were positive by PCR. Among these posi-tive samples,7 samples were taken at the first week of onset and 2 were taken on the first day. It suggests that PCR is a rapid diagnostic method to Herpes simplex encephalits.
