摘要
用MTT法检测DNR及HHT对白血病细胞的毒性作用,并观察中药汉防己甲素(tetrandrine,TTD)对两种化疗药物细胞毒性的影响。结果TTD可明显提高DNR及HHT对耐药细胞(K562/A02,K562/HHT)的毒性作用,IC50值分别下降15.8及19.1倍(P<0.05);而TTD对K562/S敏感细胞无明显影响(P>0.05)。同时进行了异搏定的对照实验。推测TTD可能是通过抑制耐药肿瘤细胞膜上P-170糖蛋白的功能而起作用的。
Cytotoxicities of daunorubicin(DNR)and homoharringtonini(HHT) in leukemic cells were measured by MTT method.We studied the effect of Chinese medicine,tetrandrine(TTD)on the cytotoxicities of DNR and HHT. The results showed that TTD could significantly potentiate the toxicities of DNR and HHT in drug-resistant leukemic cell lines (K562/A02,K562/HHT),the IC50 values decreased 15.8 bold and 19.1 fold respectively(P<0.05);but it′s effect on drug-sensitive cell lirie(K562/S)was negligible(P>0.05).Meanwhile,we made the control experiments with verapamil(VRP).TTD may act by means of suppressing the function of the P-170 glycoprotein in drug-resistant tumor cells.
