霍乱弧菌脂多糖的提取与其生物学活性的测定 被引量:1

摘要 采用Westphal的热酚-水法及高速离心法从霍乱弧菌569B等菌株中提取脂多糖,用血凝法测其效价。并以提取的脂多糖刺激小鼠腹腔巨噬细胞诱生肿瘤坏死因子的能力测其生物学活性。实验表明霍乱弧菌569B菌株的脂多糖有较强的诱生肿瘤坏死因子的能力,而且其血凝效价也较高。 Vibrio cholerae 569B(Inaba),BH43(Ogawa)and WJ2:(Inaba)were used in this study.Lipopolysaccharide(LPS)was extracted from those bacterial walls by hot phenolwater method and purified high-speed centrifugation. The composition of LPS was examined by Folin-Lowry method and antranone method.It was demonstrated that LPS Contains no protein and glucose level of 1.92g/100ml(569B),1.82g/100ml(BH43),1.72g/100ml(WJ2).The murine perterol nacrophage of thioglycollate. A serial dilution of LPS was added to macrophage. The activity of tumor necro-factor (TNF) of macrophdge was examined with MTT method and compared effect of LPS of three vibrio cholerae. The most interesing results were that the activity of LPS of VC569B is most effect than others,as compared with hemagglutination test also. where this result isconsisted with immunological protection of vibrio cholerae. Itwds necesxity that the further study waS been made in the mice.
