冷冻保存的人类精子超微结构变化 被引量:10

摘要 利用塑料细管法对9例有生育力供精者的精液进行冷冻保存,复温后在透射电镜下观察,发现精子的超微结构发生程度不同的损伤性变化。表现为精子外膜变薄、扭曲变形或破损,顶体肿胀,线粒体透亮度增加,嵴内间隙增宽,但顶体内膜和顶体中纬段仍保持完整。本文指出:冷冻复温导致精子外膜和顶体膜的通透性增加或破损,引起精子内容物漏失和线粒体功能障碍,精子能量代谢降低或停止,因而精子的活动率和运动速度在复温后低于新鲜精子。但冷冻复温并不影响精子对宫颈粘液和卵子透明带的穿透能力。 Semen was collected from 9 fertile male donors. Each sample was cryopreserved using France straw,and thawed in a water bath at 37℃. Electron microscopical studies showed considerable ultrastructural changes as compared with fresh semen. The anterior segment of the cell and acrosomal menbrane became increasingly swollen , tortuous , and discontinuous. The content was thined out whereas the inner acrosomal membrane equatorial segment usually preserved. The mitochondrial matrix got lighter with the swelling of the intracristal spaces. It is suggested that freezing-thawing make the plasma and acrosome membrane rupture. Some contents in sperm leak out and the energy metabolism decline. Therefore, the motility and velocity of cryopreserved sperm are lower than that of fresh sperm , but freezing-thawing doesn't interfere with the penetration of the sperm to the cervical mucus and the zona pelluci-da of the egg.
