反相高压液相色谱法测定植酸研究 被引量:6

摘要 本文建立了反相高压液相色谱测定植酸的方法。采用ZorboxC8色谱柱和0.025mol/L磷酸二氢钾为流动相,流速为0.5ml/min,用5%三氯乙酸抽提样品,抽提液过滤后直接注入色谱柱测定,用紫外检测器检测植酸,检测波长为254nm,在10~80μg范围内,线性回归方程为Y=0.0317+0.009628X(r=0.9995)。本方法应用于食品中植酸测定,与分光光度法所测结果进行比较,差异无显著性(P>0.05)。 A reversed-phase high performance liquid chromatographic method for determination of phytic acid is described.After being extracted with 5% TCA,the samples were directly injected into Zorbax C8 column.0.025%mol/L KH2PO4 was used as mobile phase with a flow rate of 0.5ml/min.Bhytic acid was determined with UV detector at 254nm.Over the weight range of 10~80μm,the linear regression equation was:Y=0.0317+0.009628Xr=0.9995).The method has been successfully employed to determine the phytic acid in different foods,The results are satisfactory in comparison with those obtained with colorimetry(P>0.05),
