摘要
本文研究并开发了一种固定边界的制备型自由流动电泳分离蛋白质的技术。以微孔滤膜作为液流的固定边界,在电场的作用下,根据蛋白质所带电荷的不同,可使目标蛋白或杂蛋白从一股液流电迁移至另一股液流。对该分离过程进行了理论分析,得出了以有效电泳迁移率为参数的数学模型。以牛血清白蛋白和牛血红蛋白为分离对象,确定了电场强度、蛋白质浓度和电泳时间等因素对分离过程的影响。
A technique of fixed boundary free flow electrophoresis for preparativeseparation of proteins has been developed in this paper.Microporous membrane acts as afixed boundary of streams. The target protein or the contaminants can be electrophoreticallytransported from one stream to another based on the electric properties of the proteins.Theoretical analysis has been done on this electrophoretic separation process, leading to amathematical model in terms of effective electrophoretic mobility. The effects of electric fieldstrength, protein concentration and electrophoretic time on the separation of bovine serumalbumin and hemoglobin are experimentally determined.
