MPTP对DA、GSH和LPO浓度的影响及Deprenyl的干预

摘要 运用高效液相色谱-电化学(HPLC-EC)法、二硫双硝基苯甲酸(DTNB)比色法和硫代巴比妥酸(TBA)反应法分别检测多巴胺(DA)、谷胱甘肽(GSH)和丙二醛(MDA)含量,研究神经毒素甲基-苯基-四氢吡啶(MPTP)对这些物质含量的影响,以及单胺氧化酶B抑制剂deprenyl对MPTP的干预作用。结果表明,MPTP具有显著降低DA、GSH含量,以及增加MDA含量的作用,deprenyl可完全抑制MPTP的上述作用。 The effects of methyl-phenyl-tetrahydropyridine (MPTP) on the levels of dopamine(DA), glutathione (GSH) and malonaldehyde (MDA)were studied.Following MPTP intoxication, the contents of DA and GSH were remarkably reduced as showen by high performance liquid chromatography with electrochemical detection and dithionitrobenzoic acid (DTNB) determination, while the concentration of MDA was elevated by thiobarbituric acid (TBA) reaction. Deprenyl,a B-type monoamine oxidase inhibitor, can completely prevent against the neurotoxicity of MPTP.
