摘要
介绍一种改良的子宫内膜抗原提取方法,经SDS-PAGE及Westernblot分析证实,产物中存在多条蛋白区带,分子量分别为18.42,54,71,82及91KD,另有三条大于94KD,其中42、82、91KD及3条大于94KD的区带可和抗子宫内膜抗体发生特异反应。并在此基础上建立了抗子宫内膜抗体的ELISA测定方法,可用于人体液抗子宫内膜抗体的检测。该法灵敏度高,特异性好,方法稳定,操作简便。临床应用表明,56例内异症患者血清抗子宫内膜抗体(IgG)阳性率为66.1%,明显高于非内异症患者(其它妇科疾患)和正常人(P<0.05)。
A Modified isolation of endometrail antigens method from the human endometrium tissues has been developed. The procedures involve collagense digestion of the endometrial tissues and purification through Sephadex G-100. There are several bands in the product, include 18, 42, 54, 71, 82, 91 KD and three bands lager than 94 KD indentified by SDS-PAGE.It has also been proved by Western blot, the 42, 82, 91 KD and three bands lager than 94 KD can react specially with anti-endometrial antibodies.The ELISA method has been estabilished,and can be used for detecting anti-endomitrial antibodies in human body fluids. Serum specimens from patients with endometriosis (n=56) and patients with non-endometriosis and normal persons (n=18) were tested for the presence of anti-endometrial antibody. The serum levels of anti-endometrial antibodies and significantlly different between the patients with endometriosis (n=56)and non-endometriosis (n=75) as well as the normal group (n=18) (P<0.05).
