摘要
采用流式细胞技术观察不同浓度的汉防已甲素(Tet)对大鼠RBL肝细胞生长增殖的影响。结果表明,随着Tet浓度的增加(10~60μg/ml),G2→S期的进程加速,且S期DNA含量及G1和G2期蛋白质含量增加呈一定的量效关系。经加入外源性Ca ̄(2+)及ATP证实Tet促进肝细胞生长增殖的作用与阻断Ca ̄(2+)内流无关。提示具有钙通道阻断作用的Tet可促进肝细胞生长增殖。
The effects of different doses of tetrandrine (Tet)on growth and proliferation of cultured RBL Iiver cells were observed by FCM. The results showed that Tet can accelerate G1→S phase: DNA contents of S phase and protein contents of G_1 and G_2 were increased with increasing doses of Tet (10~60μg/ml), there is relation between doses and effects.The results further demonstrated that these actions of Tet have no corresponce with obstruction Ca ̄ (2+) influx by adding extra Ca ̄(2+) and ATP. We conclude that Tet, Having Ca ̄(2+) antagonist actions, can promote growth and proliferation of RBL Iiver cells.
