57例小儿支原体肺炎PCR检测及其意义

摘要 对57例小儿支原体肺炎鼻咽分泌物中肺炎支原体DNA进行PCR检测。采用快速制备DNA模板的方法,简化PCR实验过程,结果42例阳性,阳性率73.7%,而用支原体培养分离方法,阳性率为55.3%(x2检验P<0.05),相差显著。对其中13例PCR阳性患者经红霉素静脉滴注治疗10~14d后,再经PCR检测,8例转阴性,5例仍为阳性。因此该方法既保持了PCR的敏感性和特异性,又简化过程,可用于常规检测肺炎支原体,有助于支原体肺炎的早期诊断、早期治疗及疗效评价。 Mycoplasma DNA in nasopharynx secretion of fifty-seven pediatric mycoplasma cases is detected by using PCR quick DNA sample preparation method and simple PCR processes. The result is positive in forty-two cases by using PCR method with positive rate of 73.7%, while positive rate is only 55.3% by using mycoplasma culture method (P<0.05). Thirteen patients with PCR positive results are treated by venoclysis Erythromycin for 10~14 days and mycoplasma DNA is detected by PCR again,among them eight cases turn negative, and five cases still remain positive. So this method is not only more sensitive and reliable than conventional culture techniques,but also simplifies PCR processes. It can be used as routine mycoplasma pneumonia test,and helps to give early diagnosis, early treatment and curative effect evaluation.
